چکیده

It was the purpose of this investigation to study the reaction between alloxan and the sulfhydryl group of cysteine and compounds containing cysteine. Alloxan injected into animals produced diabetes (1). Cysteine or glutathione injected prior to alloxan, but not after alloxan, protected animals from diabetes (2). On the basis of these data it was postulated that alloxan produced diabetes by the inactivation of essential sulfhydryl enzymes. Since the action of alloxan was not affected by cysteine or glutathione given after the injection of alloxan, it seemed probable that inactivation would involve combination with an essential sulfhydryl group rather than oxidation to the disulfide linkage, since the latter would be a reversible change. Some workers (3,4) believed that the main action of alloxan on sulfhydryl groups was oxidation. Therefore, more data on the reaction of alloxan with cysteine and its derivatives were desirable. Since the compounds involved in this study were unstable, it was thought that the best approach would be to study the changes in the ultraviolet absorption spectrum of alloxan as it was mixed with cysteine and its derivatives.

The Diabetogenic Effect of Dehydroascorbic

The chemical properties of these two substances are strikingly similar. Both compounds are irreversibly decomposed under biological conditions. Alloxan has a half life of 1 minute (1) and dehydroascorbic acid has a half life of a few minutes (2). Alloxan is readily reduced to dialuric acid (3) with which it forms alloxantin (3). Dehydroascorbic acid is readily reduced to ascorbic acid (4) and a...

Alloxan and dialuric acid; their stabilities and ultraviolet absorption spectra.

It was the purpose of this investigation to determine the ultraviolet absorption spectra of alloxan and dialuric acid. Previous investigators (l-3) reported that these substances had absorption spectra without definite maxima. However, since preliminary studies indicated that maxima were present under certain conditions, it was thought that a more careful investigation should be made. Although ...

Mechanistic aspects of alloxan diabetogenic activity: a key role of keto-enol inversion of dialuric acid on ionization.

The inversion of the keto-enol stability order of dialuric acid on ionization was calculated and verified experimentally. The radical cations in both forms were characterized. The spectrum of the keto form was observed upon direct ionization of dialuric acid under matrix conditions, whereas the enol form was formed upon a sequential electron-proton-proton attachment to alloxan under acidic aque...

The mechanism of alloxan and streptozotocin action in B cells of the rat pancreas.

Alloxan and streptozotocin are widely used to induce experimental diabetes in animals. The mechanism of their action in B cells of the pancreas has been intensively investigated and now is quite well understood. The cytotoxic action of both these diabetogenic agents is mediated by reactive oxygen species, however, the source of their generation is different in the case of alloxan and streptozot...
